The well shape decides which plate to pick. Flat bottom wells give an even liquid layer that a plate reader can measure optically, so they are the usual choice when colour development in an ELISA is read as absorbance. U bottom wells collect small volumes at the centre, which helps with mixing, dilution series, and tests where a settled button or even film is judged by eye.
