Students add a few drops of Molisch reagent to the test solution in a clean test tube, mix gently, and then let concentrated sulphuric acid run slowly down the inside of the tilted tube so that it settles as a separate layer at the bottom. The acid dehydrates any carbohydrate present, and the products react with the reagent to form a violet ring at the junction of the two layers. Sugars such as glucose and sucrose and polysaccharides such as starch all give a positive result, which is why the test is used first, before more specific tests for reducing sugars or starch.
Because concentrated acid is involved, the acid layering step should be demonstrated by the teacher or done by students wearing goggles and gloves, with the tube held away from the face. Record the result against a blank of distilled water for comparison.
