What Is a Phase Contrast Attachment Used For?
Living cells, protozoa, cheek epithelium and many bacteria are almost transparent, so under ordinary bright field they look faint unless they are stained — and staining usually kills them. Phase contrast turns the tiny delays that light suffers passing through different parts of a cell into differences of brightness, so nuclei, membranes and organelles appear with clear contrast in living, unstained material.
To set up, turn the condenser turret to the annulus that matches the phase objective in use, then look through the centering telescope in place of an eyepiece and centre the bright ring on the dark phase ring. Place the green filter in the light path for a crisper image. Turning the turret to the zero position returns the microscope to normal bright-field work for stained slides.
