What Is Immersion Oil Used For?
At the highest magnification, light leaving the slide bends sharply as it passes from glass into air, and much of it misses the objective. Filling that air gap with a transparent oil of high refractive index keeps the light path continuous, so the objective gathers more light and resolves finer detail. That is why Gram-stained bacteria, blood cells and chromosome spreads are examined under oil in microbiology lab practicals.
Focus the specimen first with the next lower dry objective, swing it aside, and place one small drop of oil on the cover glass over the area of interest. Rotate the oil immersion objective into the drop, focus with the fine control only, and avoid swinging a dry objective back through the oil. After the session, wipe the oil from the objective and the slide with lens tissue.
